Applications of differential scanning calorimetry in the development of liquid formulations for protein biopharmaceuticals

چکیده

There is a strong demand on the market for protein biopharmaceuticals that are formulated as liquids. They are easier for the physicians to handle and cheaper for the biopharmaceutical industry to produce as compared to lyophilisates, as no lyophilization step is needed. Optimally, the biopharmaceutical drug product (DP) should have a shelflife of at least two years. Recombinant proteins, however, have limited chemical or physical stability in liquid formulations. Having a multitude of biotechnology products currently in development, it is necessary to use technologies like differential scanning calorimetry (DSC) to efficiently deliver optimized liquid formulations. This application note presents the function that DSC has in the process of liquid formulation development.

Challenges to Improve the Stability and Efficacy of an Intravesical BCG Product

The aim of this investigation was to improve the storage stability and survival rate of intravesical BCG product, manufactured with an attenuated strain of Mycobacterium bovis (Pasteur strain 1173P2 of BCG) in the presence of sodium glutamate. Formulations with various concentrations of trehalose (a known protectant) were developed as liquid and lyophilized forms. Formulations were evaluated by...

Applications of isothermal titration calorimetry and differential scanning calorimetry in biopharmaceutical formulation development

One key factor in stabilization of protein drugs in liquid formulations is the choice of appropriate excipients at optimum concentrations that provide extended shelf life while also ensuring highest safety to the patient. Although general principles of stabilization have emerged from the literature over the past decade, the mechanisms by which excipients can improve the stability of a protein d...

Evaluation and Characterization of Free and Immobilized Acethylcholinesterase with Fluorescent Probe, Differential Scanning Calorimetry and Docking

Acetylcholinesterase (AChE) enzyme which catalyses the hydrolysis of choline esters, such as acetylcholine, is very important in nerve function. Previous structural studies showed the possible amyloid fibril formation on the AChE. Therefore it is important to understand interaction of ligands to prevent the formation of  amyloid fibrils. The purpose of the present study was to  char...

Improving the dissolution properties of spironolactone using liquisolid technique

In this study the effect of liquisolid technique on the dissolution profile of spironolactone was evaluated. Different formulations of spironolactone liquisolid compacts were prepared using various amounts of non-volatile vehicles (Poly ethylene glycol 400 and glycerin). The ratio of microcrystalline cellulose (as carrier) to silica (as coating powder material) was 20 for all formulations. Afte...
